The distance between chromatographic peak centers cannot establish resolution without the corresponding width basis.
Name the width convention
A baseline-width definition of chromatographic peak resolution writes Rs = 2(tR2 - tR1)/(wb1 + wb2). Retention times and both baseline widths use the same time unit. The peak-center gap and peak widths carry different information. A wide pair can overlap more than a narrow pair with identical center locations. Reporting only the retention-time difference omits the quantities needed by this defined resolution calculation.
Compare two original peak pairs
Let fictional peaks have centers at 5 and 6 minutes. Assign baseline widths 0.4 and 0.6 minute. The gap is 1 minute and Rs = 2 times 1 divided by 1.0 = 2. In a second invented pair at the same centers, assign both widths 1 minute; Rs becomes 1.
The unchanged gap does not preserve the calculated resolution. Nor does the calculation prove that actual asymmetric or unresolved peaks can be measured as these simple rows suppose. The shapes and widths in this worksheet are stipulated inputs, not observations from an instrument.
Retain the width measurements
Record each center, each width, the method of obtaining baseline width, the formula and the result. Link the fields to the source trace so another reviewer can check which peak and width each value represents. Keep alternative half-height measurements identified separately.
Before making an acceptance decision, establish the method-specific criterion and whether the observed peaks support the chosen width definition. The invented Rs values show arithmetic only. They provide neither a universal pass level nor proof of identity, impurity measurement or adequate performance for a particular pharmaceutical assay.
