Define the Interface or Transport Basis

Merck explains analyte binding to filtration devices, while its product-recovery technical brief identifies product remaining in system hold-up. Liquid-volume retention and surface-associated loss require different evidence even when both reduce the collected product amount.

Review a Hypothetical Comparison

For an invented closed comparison, start with 100 mL containing 2 mg/mL of solute: 200 mg total. Suppose the system retains 8 mL at an independently measured 2 mg/mL, accounting for 16 mg. The collected 92 mL at 1.9 mg/mL contains 174.8 mg. The unaccounted 9.2 mg cannot automatically be assigned to adsorption: sampling, concentration gradients, degradation or another unmeasured route also need checking. The hold-up calculation identifies one known contribution.

Keep the Evidence with the Decision

Draw the collected and retained liquid compartments and pair their volumes with actual concentrations. Keep rinses, sampling withdrawals and other outputs in the mass balance. Request a suitable independent method if surface binding is the proposed explanation. Do not call volume recovery a solute-recovery result when concentrations differ.
